Primer set for PCR detection of mycoplasma contamination in cell culture
Mycoplasma Primer Set A: The outer and inner primers of Primer Pair A are designed to detect M. orale, M. salivarium, M. hominis, M. hyorhinis, M. fermentans, M. gallisepticum, and A. laidlawii. These primers target the conserved region of the 16S rRNA small subunit (NCBI Accession # M24659). The outer amplicon is 569 bp and the inner amplicon is 288 bp. The primer binding sites for the Inner Primer Pair are within the amplicon produced by the Outer Primer Pair.
Mycoplasma Primer Set B: The outer and inner primers of Primer Pair B are designed to detect M. genitalium, M. hyponeumoniae and M. flocculare. These primers target the conserved region of the 16S rRNA gene (NCBI Accession# X77334). The outer amplicon is 698 bp and the inner amplicon is 300 bp. The primer binding sites for the Inner Primer Pair are within the amplicon produced by the Outer Primer Pair.
FORMAT: This product consists of four vials of primers, each containing 0.5 mL of Forward and Reverse oligonucleotide primers. Each primer is at a concentration of 5 µM in 10 mM Tris-HCl pH 8/0.1 mM EDTA.
STORAGE CONDITIONS: Store at -20ºC
SHELF LIFE: 3 years when stored under optimal conditions
RECOMMENDATIONS FOR USE: Use 4 µL of each primer set per 40 µL PCR reaction, or 5 µL per 50 µL PCR reaction. The primers were designed to have standard annealing temperatures (55°C).
